Microbial Expressed Antibodies Custom Service
The development of antibody therapeutic has made significant progress over the past years. Initially, the major focus has been on the generation of antibodies with glycosylation patterns similar to naturally immunoglobulin Gs (IgGs) using mammalian cells. However, the disadvantage of inconvenient and time-consuming process and the risk of viral contamination in turn has fueled interest in other production systems such as microbial hosts.
Increasingly successes in expression of antibody-related products demonstrate the importance of microbial host application. The use of microbial cells such as yeast and bacteria for antibody customization has many advantages, such as short production cycle, low cost, and simple genetic manipulation. A number of bacteria have been used for the production of recombinant proteins, such as Escherichia coli, Corynebacterium glutamicum, Pseudomonas putida, and Bacillus megaterium. Of these, E. coli is the most widely used bacteria host.
Lots of advances have been made for enhancing antibody productivity in bacteria, including translation region initiation (TIR) optimization, engineering of secretion pathways, and co-expression of molecular chaperones. Bacterial expression systems are ideal for expressing single chain antibody scFvs, Fabs and artificial minibodies. But production of full-length antibodies (IgG) in E. coli remains a challenge.
Eukaryotic cells such as yeasts offer additional advantages of post-translational modifications, which facilitates proper protein folding. Pichia pastoris, Saccharomyces cerevisiae, Yarrowia lipolytica, and Schizosaccharomyces pombe have been used for antibody production.
