Chlorine (Available) in Disinfectants Germicidal Equivalent, Disinfectants Testing

This test method is suitable for evaluating sanitizing rinses for previously cleaned non-porous surfaces, especially for the efficacy test of halogens such as chlorine, bromine, iodine. Through this test method, the amount of the test product with equivalent performance to the known concentration of sodium hypochlorite can be determined.

Test Process

• Prepare the test culture.
• Prepare NaOCl solutions containing 200, 100, and 50 ppm (µg/mL) available Cl respectively. Water bath at 20°C.
• Prepare the disinfectant product to be tested by diluting with sterile H₂O. Water bath at 20°C.
• Add a quantitative test culture to the standard solution containing 200 ppm available Cl, shake and water bath for 1 min, then transfer to the subculture tube.
• After 30 seconds, add the test culture again, shake, and after a water bath for 1 min, perform the second subculture. After 30 seconds, add the test culture for the third time.
• Repeat the steps of adding test culture and subculturing until a total of 10 test cultures are added.
• Repeat the operation in the same way for NaOCl standard solutions containing 100 and 50 ppm available Cl respectively.
• For the disinfectant product solution to be tested, perform the test in the same way as above.
• All test tubes were incubated at 37°C for 48 hours, and then observe and record the growth status.
• Determine the product concentration equivalent to the standard chlorine solution by comparing the culture results of the test product solutions and the NaOCl standard solutions.
