Multilocus sequence typing (MLST) is a typing method derived from multilocus enzyme electrophoresis (MLEE), which is widely used in the typing of bacteria, epidemiology and microbial evolutiology. MLST is a technology for strain typing by directly determining the nucleotide sequence of multiple housekeeping genes, which can classify the same species of bacteria into more subtypes.
There are some housekeeping genes in bacteria, and these gene fragments are of different lengths and have different polymorphisms in different strains. MLST is a bacterial typing method based on the comparison of nucleotide sequences. It usually compares the polymorphism of the nucleotide sequence of the housekeeping genes within the bacteria (generally 7 housekeeping genes are selected). Design primers for housekeeping genes, and amplify the nucleotide sequence of housekeeping genes in bacteria by PCR. By sequencing the amplified product and comparing it with the corresponding gene in the database, an allelic profile can be obtained and the sequence type (ST) of the strain can be determined. Each ST represents a set of individual nucleotide sequence information, and the correlation between strains can be found by comparing the ST of the strains, so as to achieve the purpose of strain typing.
The use of MLST technology can realize the ST analysis of strains, as well as phylogenetic tree analysis and clone complex analysis. MLST can be used for typing up to 100 kinds of bacteria, applicable strains include Bacillus cereus, Bacillus licheniformis, Listeria monocytogenes, Propionibacterium acnes, Salmonella enterica, Campylobacter spp, Cronobacter spp, Neisseria meningitidis, Staphylococcus aureus, Streptococcus pyogenes, Candida albicans, Escherichia coli, etc.
