Pulsed-Field Gel Electrophoresis (PFGE)

Pulsed-Field Gel Electrophoresis (PFGE) is a mature molecular biological bacterial typing technology method. This method has the advantages of good typeability, good reproducibility, strong discriminating power, and has standardized technical solutions. It is considered as the "gold standard" for bacterial typing. PFGE separates DNA molecules of different sizes by changing the direction of the electric field and the appropriate pulse time (suitable for the separation of large molecular DNA, which can separate DNA molecules up to 10Mb). The use of PFGE to study bacterial strains is of great significance in tracing the source of infection and investigating the route of transmission in epidemiology.

Principle of PFGE

Pulsed-Field Gel Electrophoresis (PFGE)

PFGE changes the swimming direction of the DNA molecules embedded in the agarose gel through the continuous change of the electric field. Small DNA molecules change faster than large ones, so that the gel shows an electrophoretic band pattern according to the size of the chromosome. In bacterial typing, the bacterial chromosomal DNA is embedded in a gel block, hydrolyzed with protease, and restriction endonucleases with rare enzyme cutting sites are used to digest the bacterial genomic DNA. The obtained DNA fragments are separated in an agarose gel applied with a pulsed electric field to produce different DNA bands. The density of DNA bands reflects the content of DNA in bacteria and the size of DNA molecules to a certain extent, and finally achieves the purpose of strain typing.
