Germicidal Spray Products Test

Summary of Germicidal Spray Products Test

The AOAC germicidal spray products test (AOAC official method 961.02) was originally developed in 1961 and is one of the antimicrobial pesticide registration test methods designated by the U.S. Environmental Protection Agency (EPA) to verify bactericidal claims of disinfectant spray products (such as trigger sprays and aerosols). It is a standard method for evaluating the efficacy of liquid disinfectants on hard, non-porous surfaces. This method is particularly suitable for disinfectants that do not require dilution and are designed to be used with aerosols or trigger sprays. It should be noted that this method is semi-quantitative, and it relies on a series of qualitative observations (test tubes change from clear to turbid due to microbial growth).

A revised version of this method was published in the AOAC Official Methods of Analysis in 2009. The revised version improves the clarity of the method and eliminates extensive cross-referencing to other methods, thus creating a stand-alone program. However, the germicidal spray products test method still lacks a standard procedure for enumeration of the test microbe on untreated dry inoculated glass slides carriers, and associated validity requirements (that is, minimum and maximum levels of the test microbe), resulting in that the relevant data may not be repeated within a laboratory or between laboratories.

Therefore, Creative BioMart Microbe improved the program to solve these problems, that is, to establish a program to enumerate bacteria from the inoculated control carriers, the minimum and maximum control carrier count range to verify the efficacy results, and the use period for inoculated carriers. These improvements will further standardize the germicidal spray products test and increase confidence in the efficacy results of products within and between laboratories.

Organisms that can be tested by this method:

  • Salmonella enterica
  • Staphylococcus aureus
  • Pseudomonas aeruginosa
  • Trichophyton mentactrophytes

Advantages of the AOAC Germicidal Spray Products Test

  • The AOAC germicidal spray products test is an antimicrobial pesticide registration test method designated by the EPA for data submission in conjunction with registration of spray (aerosol/trigger-spray) disinfectants sold in the USA.
  • The AOAC germicidal spray products test is a "high level" test for spray disinfectants. They must have significant biocidal activity within a relatively short (generally less than 10 minutes) time frame to pass the test.

Disadvantages of the AOAC Germicidal Spray Products Test

  • The AOAC germicidal spray products test is fundamentally similar to the AOAC use-dilution test, and is variable on the basis of statistics alone.
  • The specified 10-minute “wet dwell” time is relatively long and unrealistic in real life, and is not adhered to by most consumers of spray disinfectant products.
