Microbial Enumeration Test

Microbial enumeration refers to the counting of microorganisms other than viruses. The counting of microorganisms is the first step in performing various tests on microorganisms. The detection of microorganisms is of great importance both in theoretical research and in production practice. Creative BioMart Microbe has taken advantage of the platform to develop different counting methods for different microorganisms.

Microbial Enumeration Test

Total cell count can be determined electronically or by direct counting. Liquid samples containing low cell concentration can be determined by membrane filtration and staining the cells on the membrane, then counting the number of live cells under a microscope. Certain types of cells, which tend to be clumped or chained, cannot be counted accurately by the above method, but can be counted on the basis of "colony-forming unit / volume". Microbial enumeration tests are performed to determine if the product is contaminated and to determine the total aerobic microbial count (TAMC) and total yeast and mold count ( TYMC).

Plate Count Test Methods

The bacteria solution to be tested was serially diluted and cultured on a solid medium plate. The number of bacteria contained in the original bacterial solution can be calculated by the number of colonies appearing on the plate. Plate count test methods can not only obtain the number of live bacteria in the bacterial liquid, but also isolate and culture the bacteria in the bacterial liquid to obtain a single clone. Plate count test methods for microbial enumeration can be divided into pour-plate method and surface-spread method.

Membrane Filtration Test Method

Filter a certain volume of bacteria-containing sample with a special filter membrane, and then stain it with acridine orange. When the fluorescence of the cells is observed under a UV microscope, live cells will fluoresce orange, while dead cells fluoresce green.

Hemocytometer Method

This method is simple, intuitive and fast, but it is only suitable for counting spores produced by microorganisms in single-cell state or filamentous microorganisms, and the result obtained is the total number of bacteria including dead cells.

Stain Counting Method

This method makes up for the shortcomings that some microorganisms are not easy to observe and count under the oil microscope, and the direct hemocytometer method cannot distinguish dead cells from living cells.

Proportional Counting Method

Mix the liquid with known concentration of particles and the bacterial liquid with a cell concentration to be tested uniformly in a certain proportion, and count the respective numbers in the field of view of the microscope to obtain the cell concentration of the unknown bacterial liquid.

Liquid Dilution Method

Serial ten-fold serial dilutions were made on unknown bacterial samples. Samples were inoculated from the most suitable three consecutive 10-fold dilutions, and the number of test tubes with growth in each dilution was recorded after incubation. Then check the most probably number table to get the number of bacteria contained in the bacteria sample, and calculate the viable bacteria content according to the dilution factor of the sample. This method is commonly used in the detection of microorganisms in food.

Reagent Paper Method

The reagent paper method was developed based on the plate count method. Reagent paper method has the advantages of fast and accurate counting.
