qPCR technology can be used for absolute quantification of bacterial content and is the gold standard for absolute quantification of the abundance of microorganisms. However, the use of qPCR for quantitative analysis of bacteria has the disadvantage of unstable experimental results. In addition, qPCR experiments involve primer design and have higher requirements for primer specificity. The microbial community has the characteristics of diverse species, and this will increase the difficulty of using qPCR technology to analyze the microbial community. Another challenge for the analysis of bacteria in environmental samples by qPCR is that the samples contain humic acid, which can affect the accuracy of qPCR results.
