Repetitive Element Palindromic PCR (rep-PCR)
Repetitive Element Palindromic PCR (rep-PCR) is a PCR-based method for bacterial genotyping. Using the difference of repeating sequence units between different strains, the DNA fingerprints of bacterial strains can be distinguished, so as to achieve the purpose of strain typing.
Principle of Repetitive Element Palindromic PCR (rep-PCR)
Repetitive sequence units are not only present in the genome of eukaryotes, but also contain multiple scattered repetitive sequences in the bacterial genome. The number and distribution of these repeating sequence units among different strains have their own characteristics. These sequences are highly conserved in the evolution process, distributed at different sites in the genome and separated by different distances, and there are differences at the strain and species level. Using bacterial genomic DNA as a template and using primers complementary to these repetitive sequences to amplify different regions between these sequences, DNA fragments of different sizes can be obtained. Amplified products can be separated by gel electrophoresis of multiple amplicons of different sizes. Microbial DNA fingerprints can be obtained by rep-PCR. These unique DNA fingerprints define the chromosomes of different bacteria, so this technology can be used for strain typing.
