Genome Analysis for Drug and Molecule Development

Genomics, particularly high-throughput sequencing and characterization of expressed human genes, has been used as a tool for accelerating drug development. Pharmacogenomics is the application of genomic technologies such as gene sequencing, statistical genetics, and gene expression analysis to drugs in clinical development and on market.

Three genome analysis techniques, genome-wide association studies (GWAS), exome sequencing, and whole genome sequencing, are currently use in drug development. GWAS are mainly applied for advancing the understanding of the biological basis of common disorders such as cardiovascular disease, diabetes, infectious diseases, inflammatory and autoimmune disorders. Exome analysis is able to identify rare and common coding variants, which allows for the assessment of the likelihood of functional impact and predicted deleteriousness by various predictive metrics.. Whole genome sequencing is of particular interest for the study of rare genetic disorders on clinical grounds.

Transcriptional profiling of cells and tissues is perhaps the most common technologies, that is used in mapping responses to compounds, the interrogation of tissues and cells for the expression of a target of interest, assisting with the identification of causal variants associated with clinical phenotypes. Moreover, transcriptomics is also an important tool for the discovery of biomarkers for patient stratification in clinical trials.
