Generally, the minimum inhibitory concentration (MIC) assay is used to test the efficacy of antibiotics due to its rapid and reproducible advantages. Although the traditional MIC is the most commonly used method, this method can only test the efficacy of antibiotics against free-floating, planktonic bacteria and fungi, but cannot effectively test the efficacy of antibiotics against adherent biofilms. Biofilm is a matrix formed by extracellular polysaccharides and extracellular polymers, which is a matrix formed when microbes adhere to the surface. Biofilm is a natural protective barrier that can effectively prevent antibiotics from killing enclosed bacteria and fungi. Therefore, antibiotics cannot successfully penetrate the biofilm, resulting in antibiotic resistance.

Biofilms exist wherever these microbes live, so the antibiotics, preservatives or disinfectants used must be effective for the biofilms. In order to test this resistance and gain insight into biofilms, biofilm eradication testing is required.
