LAL Assay
Limulus amebocyte lysate (LAL) is an extract of blood cells (amoebocytes) from the horseshoe crab Limulus polyphemus that can undergo a clot reaction with endotoxin. The LAL assay is the gold standard for endotoxin testing. LAL assays include gel methods and photometric methods, the latter of which in turn includes chromogenic and turbidimetric methods.
Gel colt LAL assay utilizes the clot reaction of LAL reagent and endotoxin to perform qualitative or semi-quantitative detection of endotoxin in a sample. The gel LAL test has the advantage of being simple to operate by observing the presence or absence of gel formation as the endpoint of the reaction. The turbidimetric LAL assay utilizes the change of turbidity during the reaction between LAL reagent and endotoxin to determine the content of endotoxin, which can realize the accurate quantification of endotoxin, and has the advantages of simple operation and high sensitivity. The chromogenic LAL assay utilizes the coagulase produced during the reaction between LAL reagent and endotoxin to develop the color of a specific substrate to determine the endotoxin content. It also has the advantages of simple operation and high sensitivity.
