Product Details
Terrific Broth (TB), Modified is a liquid medium used with Glycerol in cultivating recombinant strains of Escherichia coli. Although the bacteria are slower growing in this medium, the intention of Terrific Broth, Modified is to improve plasmid yields and cell density. Tryptone provides nitrogenous compounds and minerals, while Yeast Extract is a source of B-complex vitamins. Mono- and Dipotassium Phosphates serve as pH buffers.
Product Ingredients
Tryptone (12.0g/L), Potassium Phosphate, Monobasic (2.2g/L), Potassium Phosphate, Dibasic (9.4g/L), Yeast Extract (24.0g/L). Final pH: 7.4 ± 0.2 at 25°C. Grams per liter may be adjusted or formula supplemented to obtain desired performance.

Usage Method
Mix 47.6 grams of the medium and 4ml of Glycerol in a Liter of purified water until evenly dispersed. Heat with repeated stirring to dissolve completely. Distribute and autoclave at 121°C for 15 minutes.

Storage
Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original beige to yellow beige color.
