Reinforced Clostridial Medium is a nonselective medium for the cultivation, isolation and enumeration of Clostridium spp. and other anaerobes. The medium is highly nutritious and can be used in the enrichment of growth broths before subculturing clostridia to solid agar. Beef Extract and Casein Peptone provide nutrients such as amino acids, peptides, carbohydrates and vitamins. Dextrose is an energy source for the bacteria. Sodium Chloride provides essential electrolytes and maintains osmotic equilibrium. Low concentrations of Soluble Starch help detoxify metabolic by-products. L-Cysteine HCl acts as a reducing agent and Sodium Acetate as a buffer. The medium contains a small amount of Agar to concentrate CO2 while dispersing O2 and reducing substances.

Enzymatic Digest of Casein (10.0g/L), Beef Extract (10.0g/L), Dextrose (5.0g/L), Sodium Chloride (5.0g/L), Yeast Extract (3.0g/L), Sodium Acetate (3.0g/L), Soluble Starch (1.0g/L), L-Cysteine HCL (0.5g/L), Agar (0.5g/L), Final pH: 6.8 ± 0.2 at 25°C. Grams per liter may be adjusted or formula supplemented to obtain desired performance.

Usage Method
Mix 38 grams of the medium in one liter of purified water until evenly dispersed. Heat with repeated stirring to dissolve completely. Autoclave at 121°C for 15 minutes.

Storage
Store the sealed bottle containing the dehydrated medium at 2 to 30°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original light beige.
