Product Details
Rappaport-Vassiliadis R10 Broth is used for the selective enrichment of Salmonella spp. from meat products, feces, and water polluted by sewage. The medium was formulated by Rapport and modified by Vassiliadis by reducing the concentrations of Malachite Green and Magnesium Chloride. Enzymatic Digest of Casein provides amino acids, peptides, carbohydrates, vitamins and electrolytes for bacterial growth. Sodium and Magnesium Chloride supply essential electrolytes and maintain the osmotic equilibrium. Potassium Dihydrogen Phosphate serves as a pH buffer. Malachite Green specifically enriches for Salmonella, since other species are typically resistant to or inhibited by this compound.
Product Ingredients
Enzymatic Digest of Casein (4.5 g/L), Magnesium Chloride Anhydrous (13.4 g/L), Sodium Chloride (7.2 g/L), Malachite Green oxalate (0.036 g/L), Potassium Dihydrogen Phosphate (1.45 g/L). Final pH: 5.1 ± 0.2 at 25 °C. Grams per liter may be adjusted or formula supplemented to obtain desired performance.

Usage Method
Mix 26.6 grams of the medium in one liter of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave at 116°C (10 lb. pressure) for 15 minutes.

Storage
Store the sealed bottle containing the dehydrated medium at 2 to 30°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original color.
